In addition, in both patients allergic patch-test reactions to another Asteraceae species (Liatris spicata) could be demonstrated. This Asteraceae species is a native flower (Blazing Star) in the USA, but in Germany we find the flower in shops, and sometimes in parks and gardens. Genus Liatris contains about 30 species. Structure determination of pycnolide as a 2,3-seco-1(10) 4,5-germacradienolide is detailed. The previously known cytotoxic guaianolide spicatin, the antileukemic 5,10- epoxygermacranolide chapliatrin, and euparin were isolated from L tenuifolia Nutt. The cytotoxic heliangolide eleganin was isolated from L scabra (Greene) K. Schum. L earlei (Greene) K. Schum and L pauciflora Pursch gave small amounts of complex lactone mixtures. The lactone ring is trans fused. The 4,5 double-bond in the cyclodecadiene is cis oriented, while the 1,10 double-bond is trans oriented. The C-8 ester side chain contains two 5-carbon acyl units, one of which is the cis isomer of sarracinic acid. Provincialin had significant cytotoxic activity (ED(50) of 3.5 microg/ml) against KB carcinoma cells of the human nasopharynx. The isolation and structural elucidation of the active constituent, liatrin, are now reported. The compound possesses an unusual germacranolide cis,cis-diene structure; it showed significant antileukemic activity against P388 lymphocytic leukemia in mice and cytotoxicity (ED(50)) against KB cell culture at 1.5 microg/ml.
